چکیده
OBJECTIVE To develop an efficient infusion technique for human spermatogonial stem cell transplantation. DESIGN A mixture with ultrasonic contrast, computerized tomography (CT) contrast, and Chinese ink was injected into isolated human testes through different sites (the rete testis, the head of the epididymis, the deferent duct, and blind testicular infusion). Ultrasound transducer was used to visualize the injection site and to observe the flow of the mixture injected in the testes. Then, micro-CT scan was used to construct three-dimensional images, allowing the calculation of the testicular volume filled by the mixture. Finally the efficiency of the infusion was evaluated on histologic sections. SETTING Research laboratory. PATIENT(S) Cadaver testes obtained from autopsied bodies at the department of pathology. INTERVENTION(S) Ultrasound-guided infusion of contrast liquid. MAIN OUTCOME MEASURE(S) Contrast liquid-filled testis volume and presence of ink in seminiferous tubules. RESULT(S) Ultrasonography clearly visualized the flow when seminiferous tubules were injected from the rete testis. No flow was observed when infusions were made either blindly, into the deferent duct, or into the head of the epididymis. On micro-CT no significant differences were observed between the different volumes. After rete testis infusion, ink particles were found in the lumen of the rete testis and in tubules, close and distant from the rete testis. CONCLUSION(S) A single ultrasound-guided injection of 800 μL in the rete testis may provide a promising method to transplant human spermatogonial stem cells in a clinical setting.

O-19: Proliferation of Small Population of Spermatogonial Stem Cells in Azoospermic Patients
Background: With treatment success in young boys with cancer, long-term effects of cancer treatment have found importance in pediatric oncology. Temporary or permanent infertility after treatment is an important subject in childhood and adult cancer patients which decrease quality of life. The one approach to overcome infertility in these cases is to cryopreserve small biopsy testicular tissue ...
متن کاملMelatonin improves spermatogonial stem cells transplantation efficiency in azoospermic mice
Objective(s):Transplantation quality improvement and reduction of cellular damage are important goals that are now considered by researchers. Melatonin is secreted from the pineal gland and some organs such as testes. According to beneficial effects of melatonin (such as its antioxidant and antiapoptotic properties), researchers have proposed that the use of melatonin may improve transplantatio...

Background: This study was conducted to identify and characterize repopulating spermatogonial stem cells (SSCs) in the adult human testes. Materials and Methods: Testes biopsies from obstructive azoospermic patients and normal segments of human testicular tissue were used. Flow cytometry, real time PCR and immunohistochemical analysis were performed. Purified human spermatogonia were transplant...
متن کاملAssessment of Culture Condition and In Vitro Colonization Ability of Human Spermatogonial Stem Cells: A Review Article
Spermatogenesis is a highly complex and regulated process in which germ stem cells differentiate into spermatozoa. These stem cells, called spermatogonial stem cells (SSCs), are in the base of seminiferous tubules and have the ability of self-renewal and differentiation into functional germ cells. Due to this ability, SSCs can restore spermatogenesis after testicular damage caused by cytotoxic ...
متن کاملI-4: Male Fertility Preservation Options, Current State-of-The ART and Future Implications
Background Male infertility management has made significant progress during the past three decades, especially after the introduction of intracytoplasmic sperm injection (ICSI) in 1992. However, many boys and men still suffer from primary testicular failure due to acquired or genetic causes. New and novel treatments are needed to address these issues. Spermatogenesis originates from spermatogon...
